Overview

[F-18]T808 is a small molecule diagnostic radiopharmaceutical designed as a positron emission tomography (PET) tracer for imaging tau protein aggregates in the brain. Tau aggregation is a hallmark of neurodegenerative diseases such as Alzheimer disease. [F-18]T808 exhibits high affinity and selectivity for tau aggregates over amyloid-β in vitro and demonstrates high initial brain uptake with fast washout in animal models. The tracer undergoes rapid metabolism and partial defluorination in vivo, resulting in higher bone uptake. Despite early phase clinical efforts for imaging tau pathology, clinical development for Alzheimer's disease was terminated due to limited success in later-phase studies and toxicity/metabolic concerns.[1][3]
